Abstract

AbstractIn this study the effect of 5*urosemide and prostaglan- din (PG) synthesis inhibitors ASA and Dipyrone (ÎISAI pharma- ceuticals) on urine production in dogs was investigated pharma- codynamically. The above mentioned drugs are becoming incre- singly important in the treatment of oedema of the heart, liver and kidney in veterinary medicine. For the investigation 56 dogs of mixed age and sex, between 12-29 kg were used. The animals were divided into 4 groups of 14 To the dogs used in the experiments, general an- aesthesia was carried out, a cannule was placed in the trachea, laparatomy performed and v/ith the help of a peristaltic pump, 25 ml/kg of Isotonic sodium chloride solution infused into the front leg vein to compensate for the blood loss during the operation,, To the first group, immediately after the procedure above, 0.3 ml/kg/hour of Isotonic sodium chloride solution was infused for a period of 120 minutes with the aid of the peristaltic pump. To the second group of dogs, after the initial proce- dure, 0.5/mg/kg Purosemide was infused all at once into the front leg vein with the aid of the peristaltic pump; following this a 0.3 ml/kg/hour dose of Isotonic sodium chloride solution was infused for the 120 minute period of the experiment. To the third group a 0.5 mg/kg dose of PuroserJde followed by a 25 mg/kg dose of ASA was infused in to the front97 leg vein using the peristaltic pump and following this a 0.3 ml/kg/hour dose of Isotonic sodium chloride solution was infused for 120 minutes for the duration of the expe- riment. To the fourth group a dose of 0.5mg/kg of Furosemide was followed by a dose of Bipyrone at 25 mg/kg was infused in- to the front leg vein usinfj the peristaltic pump; following this a 0.3 ml/kg/hour dote of Isotonic sodium chloride solu- tion was infused for 120 minutes for the duration of the experiment" The amount of urine formed in group 1 with Isotonic sodium-chloride solution was followed at 10 minute intervals. In the second group the amount of urine formed using Purosemide was followed at 10 minute intervals and this was used as a control for the third and fourth groups in which Purosemide and ASA and Purosemide and Dipyrone were used re- spectively. The diuretic action of Purosemide was decreased by the use of ASA in group 3 and Dipyrone in group 4 and the decrease in urine production during the experimental period was found to be statistically significant. During the 120 minute period of the experiment, the decrease in urine amounts for the third group receiving ASA Avas 15.05 % whereas this value was 18.63 % for group 4 to which Dipyrone was given These percentages were based on the values of group 2 receiving Purosemide. Thus it was de- termined that the decrease in urine production was gî^eater for the group receiving DipyroneP
